Hamilton, ON (August 31, 2011) Scientists were surprised at how fast bacteria developed resistance to the miracle antibiotic drugs when they were developed less than a century ago. Now scientists at McMaster University have found that resistance has been around for at least 30,000 years.

Research findings published today in the science journal Nature show antibiotic resistance is a natural phenomenon that predates the modern clinical antibiotic use. Principal investigators for the study are Gerry Wright, scientific director of the Michael G. DeGroote Institute for Infectious Disease Research and Hendrik Poinar, McMaster evolutionary geneticist.

"Antibiotic resistance is seen as a current problem and the fact that antibiotics are becoming less effective because of resistance spreading in hospitals is a known fact," said Wright. "The big question is where does all of this resistance come from?"

After years of studying bacterial DNA extracted from soil frozen in 30,000-year-old permafrost from the Yukon Territories, the researchers were able to develop methods to isolate DNA within McMaster's Ancient DNA Centre. Using state-of-the-art molecular biological techniques, methods were developed to tease out small stretches of ancient DNA.

Researchers discovered antibiotic resistant genes existed beside genes that encoded DNA for ancient life, such as mammoths, horse and bison as well as plants only found in that locality during the last interglacial period in the Pleistocene era, at least 30,000 years ago. They focused on a specific area of antibiotic resistance to the drug vancomycin, a significant clinical problem that emerged in 1980s and continues to be associated with outbreaks of hospital-acquired infections worldwide.

"We identified that these genes were present in the permafrost at depths consistent with the age of the other DNAs, such as the mammoth. Brian Golding of McMaster's Department of Biology showed that these were not contemporary, but formed part of the same family tree. We then recreated the gene product in the lab, purified its protein and showed that it had the same activity and structure then as it does now."

This is only the second time an ancient protein has been 'revived' in a laboratory setting.

Wright said the breakthrough will have important impact on the understanding of antibiotic resistance: "Antibiotics are part of the natural ecology of the planet so when we think that we have developed some drug that won't be susceptible to resistance or some new thing to use in medicine, we are completely kidding ourselves. These things are part of our natural world and therefore we need to be incredibly careful in how we use them. Microorganisms have figured out a way of how to get around them well before we even figured out how to use them."

Poinar says this discovery has opened doors for ancient antibiotic resistance research. "We can go back a million years in the permafrost, which is our next goal."

Funding for this project came from the Canada Research Chairs program, the Canadian Institutes of Health Research and the Natural Sciences and Engineering Research Council.

Preventing the formation of amyloid fibrils

A molecule which can stop the formation of long protein strands, known as amyloid fibrils, that cause joint pain in kidney dialysis patients has been identified by researchers at the University of Leeds.

The discovery could lead to new methods to identify drugs to prevent, treat or halt the progression of other conditions in which amyloid fibrils play a part, including Alzheimer's, Parkinson's and Type II diabetes.

The research, funded by the Biotechnology and Biological Sciences Research Council and the Wellcome Trust, is published today (August 28) in Nature Chemical Biology.

The team from Leeds' Astbury Centre for Structural Molecular Biology and Faculty of Biological Sciences found that an antibiotic known as Rifamycin SV was able to prevent the protein ?2microglobulin (?2m) from forming into fibrils. ?2m is known to accumulate in renal dialysis patients and forms fibrils within the joints, causing extreme pain and arthritis.

By using a specialised analytical technique called ion mobility spectrometry-mass spectrometry (IMS-MS), the researchers were able to see at what stage of the process Rifamycin SV prevented amyloid fibril formation. They believe the technique could enable potential drugs to be identified for the many other proteins which form amyloid fibrils, linked to a wide range of human disorders.

"Traditional drug design for diseases like Alzheimer's is incredibly difficult because the proteins you're trying to target are changing shape and structure all the time," explains University of Leeds Professor of Structural Molecular Biology, Sheena Radford. "It's like trying to consistently pick out one bead of a particular shape from box of potentially millions of similar beads. This new technique allows us to see the shape of the protein as it changes, so we can more easily identify exactly which part we need to target."

In their normal, folded state, proteins are unable to link together to form long fibrillar assemblies, but if they unfold, they expose areas where they can bind to each other. Initially they form small groups of two, three or four proteins, and then these link into long strands, which twist together to form fibrils.

Most analytical techniques can only show the mass of the protein or its make-up in terms of amino acids, neither of which changes as the protein unfolds. Others are unable to look at individual molecules within complex mixtures. However, IMS-MS can measure the mass and shape of a protein, allowing researchers to watch the unfolding process and the aggregation into small groups and then assembly into the fibril and to find which of these species is able to bind a ligand and stop the assembly process.

In the research published today, researchers found that Rifamycin SV stopped the formation of protein fibrils by binding to an unfolded protein molecule with a particular shape, enabling for the first time, an unfolded protein of a particular shape to be identified as a target for the design of new inhibitors of fibril assembly.

"We're fortunate to be one of the few universities in the UK able to use IMS-MS to study amyloid fibril formation," says Professor of Biomolecular Mass Spectrometry, Alison Ashcroft, who specialises in this type of analysis. "Although fibrils take years to develop in the body, we are able to 'grow' them in hours in the lab. By using IMS-MS to help us map exactly how they are formed, we can better understand the mechanism by which it happens and we hope find ways to stop it."

Producers for the musical "The Addams Family" said Wednesday it will end its run at the Lunt-Fontanne Theatre on Dec. 31.

When it closes, it will have had 725 performances and 34 previews. A national tour launches on Sept. 15 in New Orleans and is booked in more than 30 cities through 2012.

The musical, based on characters created by legendary cartoonist Charles Addams, currently stars Roger Rees and Brooke Shields. When it opened in 2010, it starred Nathan Lane and Bebe Neuwirth.

He?s run countless successful product launches in the New York market. But now there?s a problem with a product launch in the West Coast office, near Lake Tahoe. Upper management knows John is the man to fix it. But will he accept the assignment?

The executive team makes him an offer for a long-term, possibly permanent, relocation, and John accepts. Problem solved, right? The right man is in the right position to get the new product out without a glitch.

Now John meets with Human Resources to discuss the terms of his relocation. He?ll receive reimbursement of all moving costs, a small bonus, and the HR department will connect him with real estate agents to help him sell his home. He?s got a sizeable house in a coveted suburb ? no one thinks about how long it might take to sell a two-year-old home in an overvalued area in a rapidly depreciating market.

And frankly, the HR department doesn?t really know John, or what an important asset he is to the company. They don?t realize the executive team wants to keep him on board ? and productive ? at nearly all costs. They don?t realize how important John?s productivity is to the company?s bottom line, especially with this do-or-die product launch on the horizon.

Now John faces the most important task of his career and the burden of selling a home that is now worth less than he paid for (and less than his mortgage) ? and dropping in price each day.

John moves into temporary housing in California while his wife stays behind waiting for the kids to finish out the school year, job hunting on the Internet and trying to sell the house.?

John?s wife had planned to meet him in California within a month, but now it?s closing in on three months, with no bites on the home and a very stressed-out John splitting his attention between real estate sales and the product launch. Every other weekend, John goes home to help his wife pack some more, re-evaluate home sale plans with the real estate agent, host an open house or do work around the house to help raise the value of his home and help it ?show? better.

When he returns to work Monday after taking the red eye Sunday night, his mind still isn?t on his job or the new product. The executive team in California starts to wonder what East Coast management saw in this ?star player.? His head is barely in the project. He misses meetings, forgets important details, and the important product launch is even further behind schedule than it was before. The product is high volume, low margin, which means that every day it?s not on the market means lost revenue ? and the holiday season is coming.

When John misses a deadline to finalize an agreement with a major distributor and is locked out of their plan for the next year, the company wonders if it can recover from the estimated million loss in projected profits from the new product, the damaged relationship with the distributor, and the loss of morale across the department.

John, who was supposed to be the shining star, crash landed ? and it?s nearly impossible to put a price on the damage it did to the company, above and beyond the projected million-plus.

Let?s take the scenario one step further. After losing face, failing to sell his home, and living with six months of stress, John moves back to New York, takes his house off the market, and opens an independent consulting firm. Now the sporting goods manufacturer has to hire and train his replacement ? more lost money, time and productivity.

Libya erupts as Qaddafi's compound falls to rebels

The sprawling Bab al-Aziziya ? the symbolic heart of Muammar Qaddafi's regime ? fell to the Libyan rebels today, sparking wild celebrations in much of Libya.

In 24 hours, Muammar Qaddafi has lost the two symbols of his regime. Green Square, where he was fond of delivering rambling harangues against his domestic opponents and foreign antagonists like the US, is now being called Martyrs' Square. Bab al-Azizya, the gated residential and military district that acted as the nerve center for his regime, has been largely overrun.

Al Jazeera has been carrying hours of live footage of ecstatic Libyans stomping on a gold head of Qaddafi ripped from a monument to himself at the compound, driving their cars in circles over a massive Qaddafi-era flag in Green Square, and taking gold-plated AK-47s and other items from his compound. Sky News interviewed a man wearing a rope-thick gold chain and Qaddafi's generalissimo cap outside one of his homes. The man said he'd taken them from Qaddafi's bedroom.

Where's Qaddafi? It's anybody's guess at the moment. But for the millions of Libyans who have backed an uprising that began as a demonstration against the arrests of human rights lawyers in Benghazi on Feb. 15 and evolved into an armed, national movement to end 40 years of one-man rule, today was the day of victory. There is almost certainly some fighting ahead, and the challenges to building a new Libya ? from tribal rivalries to a damaged economy to the absence of any meaningful institutions to build on ? remain great.

Some fighting continued inside the compound ? which covers about four square miles ? into the evening, with constant sounds of gunfire and occasional larger booms from mortars, according to Reuters. During Libya's almost six-month civil war, Qaddafi's forces have frequently adopted civilian clothes and hid in homes in towns being overrun, so the potential remains for further major engagements in Tripoli.

But the rebels and their supporters aren't letting that dampen their euphoria tonight. In Benghazi, an eastern city where the National Transitional Council is based, legions poured into the streets when they heard the Bab al-Aziziya had fallen.

Men fired rifles into the air and threw hand grenades in the street in celebration. Cars sped through the streets, honking, with men hanging out and waving flags. Young men hung out of a pickup truck, chanting to Qaddafi, "you can't hide in an alley, you can't hide in a wardrobe, we're everywhere and we're coming after you" in a riff on one of Qaddafi's speeches.

Others, making fun of the speech in which he said he would find and kill the rebels "beit beit, zenga zenga" (house by house, alley by alley), chanted "house by house, wardrobe by wardrobe" ? a threat that they will not let a dictator who's also something of a clothes-horse go free.

In Tripoli, the scenes at Bab al-Aziziya were reminiscent of the way Iraqis poured through Saddam Hussein's palaces after he was defeated in 2003, or how Filipinos poured through Ferdinand Marcos's Malacanang palace after their people-power revolution succeeded in 1989.

Converting a Stovetop Recipe to a Crockpot Recipe | spanishchef.net

Crockpots vary but the low setting is typically around 100 degrees and its high setting is around 300 degrees.

Knowing this, it is fairly easy to convert most recipes for use in your slow cooker. Some adjustments to cooking times will be a judgement call on your part, but some simple guidelines should help. Just follow a few simple rules and you?re on your way.

You should decrease the liquid since it does not boil away in a crockpot.

Add cheeses and other milk product towards the end of your cooking since they tend to break down if they are cooked for too long.

Brown any ground beef before adding to the crockpot. It will taste better and have better texture this way.

Add rice and noodles at the end of your cooking as well. They will turn mushy if they are cooked too long. You can cook these separately and then combine them when it?s time to serve.

Add your spices at the end.

If a stovetop recipe calls for 15-30 minutes, the crockpot should cook it for 1 1/2 ? 2 1/2 hrs at a high temperature and 4 ? 8 hours at its low temperature setting.

If a stovetop recipe calls for 35-45 minutes, the crockpot should cook it for 3-4 hrs at a high temperature setting and 6 ? 10 hours at its low temperature setting.

If a stovetop recipe calls for 50 min to 3 hours, the crockpot should cook it for 4 ? 6 hrs at a high temperature setting and 8 ? 16 hours at its low temperature setting.

Following these guidelines will allow you to be able to convert a stovetop recipe to its delicious slow cooking counterpart!

Oil prices down as rebels capture most of Tripoli

(AP) ? Brent crude fell to near $105 a barrel Monday as Libyan rebels captured most of the country's capital, boosting hopes the OPEC nation's oil exports could resume soon.

In London, Brent crude for October delivery was down $3.22 per barrel to $105.40 on the ICE Futures exchange.

Benchmark oil for September delivery was down 25 cents to $82.01 a barrel at late afternoon Singapore time in electronic trading on the New York Mercantile Exchange. Crude fell 12 cents to settle at $82.26 on Friday.

Rebels overran most of Tripoli after a quick advance to the capital as defense of Moammar Gadhafi's regime collapsed. Gadhafi's whereabouts were unknown while two of his sons were captured by rebels.

Benchmark crude rose from $84 in February to near $115 in May as the uprising shut down Libya's oil exports of 1.5 million barrels a day. Although Libyan oil amounted to less than 2 percent of world demand, its loss affected prices because of its high quality and suitability for European refineries.

Investor fears in recent weeks that slowing economic growth in the U.S. and Europe will undermine demand for crude pushed prices back down.

Libyan oil mostly supplied Europe, and analysts expect the return of Libya crude output would impact Brent prices more than the U.S. benchmark West Texas Intermediate, or WTI.

"If the Ghadafi regime falls, Libyan oil production should gradually resume and European markets would directly benefit from that," said Victor Shum, an analyst with energy consultant Purvin & Gertz in Singapore. "Having more supply while the global economy is under threat should put downward pressure on oil prices."

Investors will be closely watching how smooth the transition to the new government will be and how fast oil production can come back on line, Shum said. Some of Libya's oil infrastructure was reported damaged during the months of fighting.

In other Nymex trading for October contracts, heating oil fell 4.7 cents at $2.87 per gallon and gasoline futures dropped 6.1 cents to $2.65 per gallon. Natural gas for September delivery sank 5.3 cents to $3.89 per 1,000 cubic feet.

Germany's Merkel renews rejection of eurobonds (AP)

BERLIN ? German Chancellor Angela Merkel is insisting that introducing eurozone-wide government bonds wouldn't solve the current debt crisis.

Merkel also said in an interview with ZDF television being broadcast Sunday she sees "nothing that points to a recession in Germany," which has Europe's biggest economy, after growth cooled in the second quarter.

Germany has led opposition to so-called eurobonds, viewed by some as a logical solution to the debt crisis that has pushed up troubled countries' borrowing costs. But critics say they would drive up financially solid nations' costs and encourage others to run up more debt.

Merkel said that "solving the current crisis will not be possible with eurobonds."
